Research has shown that mutations in the psen1 gene are common in the familial forms of Alzheimer's disease, and the Presenilin-1 protein that the gene encodes is known to be involved in the cleavage of the amyloid precursor protein. In Alzheimer's disease the amyloid precursor protein is not cleaved the normal way, and the protein accumulates in the brain damaging neuronal tracts and neurons. It is still unknown if the psen1 gene is involved in the etiology of Alzheimer's disease via another mechanism.

Professor Pertti Panula's research team at the University of Helsinki has elucidated the role of psen1 gene in the development of the neuronal histamine system and its modulation. Histamine is one of the neurotransmitters, which all are essential for cognitive functions, which in turn are impaired in Alzheimer's disease. The histamine system is altered during the progression of Alzheimer's disease.

In the study the zebrafish was used as a model organism. The rapidly developing zebrafish is suitable as a model organism, as its transparency allows researchers to study the development and function of vital organs. To study the function of psen1 gene, zebrafish that did not produce functional Presenilin-1 protein were generated. Despite the fact that the fish lacked functional Presenilin-1 they were viable and developed until adulthood.

The lack of Presenilin-1 protein induced a change in the behavior of the larval zebrafish, they did not as normal fish react to fast changes in the light intensity. "Based on previous research we know that this change in behavior is associated with lack of histamine in the brain", Panula explains.

In adulthood the motor behavior of the mutant zebrafish differed from the normal fish: the fish swam by the edges of the arena that was available and avoided the inner part. Previous studies from the group have shown that this behavioral alteration also is due to changes in the histamine system.

The researchers found that larval fish lacking Presenilin-1 protein had significantly fewer histamine neurons; in adulthood the histamine neuron number was significantly increased in these fish when compared with normal fish.

"These results reveal that the psen1 gene is a central regulator of the development of the histamine neurons and that the mutation can cause a persistent lifelong change in the neuronal histamine system. This is a very interesting finding", Panula states.

One interesting remaining question is from where the new histamine neurons arise in the brains of adult zebrafish. Are they newly differentiated stem cells or do other cells become histamine neurons? The answer is not known, but based on these results it is advisable to elucidate the role of Presenilin-1 protein in differentiation of stem cells also in the brains of mammals. "Mammals have stem cells in the hypothalamus, in the same area where the histamine neurons are located in all studied vertebrates", Panula comments.

Panula empathizes that the published study does not tell about an Alzheimer's disease mechanism in humans. The new knowledge on the function of psen1 gene and the development of the brain histamine system provided by the study is one step forward to understanding the etiology of the disease.

"We perform basic research on molecular level, from where it is a long way to treatment of human diseases. This type of research provides the findings on which the treatments are finally based", Panula says.

DOE seeking input on proposed automotive fuel cell cost and durability targets

The US Department of Energy (DOE) is issuing a Request for Information (RFI) (DE-FOA-0000855) seeking input from stakeholders on proposed cost targets for fuel cells designed for automotive applications. The purpose of this RFI is to solicit feedback from developers, manufacturers, end users, and other stakeholders on proposed cost and durability targets for automotive fuel cell systems.

The proposed targets are $40/kW in 2020 and $30/kW for the ultimate target (2030) for automotive fuel cell system cost, and 5000 hrs or 150,000 miles for durability. This RFI is not and will not lead directly to a Funding Opportunity Announcement, DOE said.

DOE is in the process of updating the fuel cell technologies Multi-Year Research, Development and Demonstration Plan (MYRDDP) and determining technology requirements for widespread commercialization of fuel cells.

As part of this effort, DOE is seeking general comments on its cost and durability targets for fuel cells for automotive applications. The targets are intended to meet end user expectations, and are not driven by the operating parameters or constraints of specific technologies.

The proposed targets included in this RFI were developed with input from the IS DRIVE Partnership (a public-private partnership includes automotive and energy companies), specifically the Fuel Cell Technical Team. DOE is requesting further stakeholder general comments are requested to refine the targets.

Comments to the RFI should address one or more of the following:

  • Appropriateness of target values

  • Comparison of targets to baseline and competing technologies (both gasoline internal combustion engine and hybrid or other advanced systems)

  • Status of fuel cell technologies in comparison to targets

  • Recommendations for testing conditions and protocols

  • Assumptions used for targets and status (e.g., platinum loading and platinum price)

Comments must be provided by no later than 11:59 PM EDT on 1 April 2013.

(AP) ? Paraguayan presidential candidate Lino Cesar Oviedo has been killed in a helicopter crash, authorities said Sunday, ending a dramatic political career that included coups and repeated attempts to lead this impoverished 6.5 million-person country.

Oviedo was returning with his bodyguard from a political rally in northern Paraguay Saturday night when his pilot encountered bad weather. All three were killed in the crash, said Johnny Villalba, a spokesman for Paraguay's airport authority.

Defense Minister Maria Liz Garcia said she traveled to the scene Sunday with Oviedo's daughter, congresswoman Fabiola Oviedo, and confirmed that the helicopter "disintegrated."

"One resident who lives near the accident scene said they heard a single explosion Saturday night," she added. "The aircraft ended up disintegrated and out of respect to the families of the victims, I won't release details about the cadavers."

The air traffic control tower in the provincial city of Concepcion received the pilot's last communication, Garcia said ? a brief message that they were changing course due to a storm at 9 p.m. local time.

Lino Oviedo, 69, was running in April's elections as leader of Paraguay's third-largest opposition party, the National Union of Ethical Citizens.

A retired general and former army chief, Oviedo had tried for years to take the helm of his nation, and not always through democratic means.

As a colonel in 1989, Oviedo had been tasked with taking prisoner none other than Alfredo Stroessner, the feared dictator who had ruled Paraguay since 1954.

That bloody military coup sent Stroessner into Brazilian exile, but did little to diminish the hold on Paraguayan politics that his Colorado Party had cemented. Oviedo's role in the ouster was rewarded with a meteoric rise through the ranks of the army.

A diminutive cavalry officer, only 5-foot-3 inches (1.62 meters) tall, Oviedo was promoted to brigadier general three months after capturing Stroessner. By 1992, he had become a division general, and then President Juan Carlos Wasmosy named him army chief.

Membership in the Colorado Party had been a requirement for any officer during the dictatorship, but Oviedo's constant involvement in party politics generated frictions with the president.

In April 1996, a short-lived coup in which Oviedo participated and other maneuverings led to his firing and forced retirement. He ran as a candidate to succeed Wasmosy, winning the Colorado Party primary ahead of the May 1998 presidential elections, but was then convicted in a military court for his role in the coup, ending his candidacy.

His would-be vice president, Raul Cubas, instead won the election and immediately ordered the release of Oviedo, despite the judiciary's rulings.

Oviedo had an irrepressible desire to govern, and quickly became known as the power behind Cubas, angering other political leaders.

Cubas' vice president, Luis Maria Argana, was assassinated in 1999, and the slain man's relatives and followers accused Oviedo of being the mastermind. Cubas resigned in the resulting turmoil and Oviedo fled the country and renounced his Colorado Party membership, founding the UNACE party.

Facing Paraguayan arrest orders in the Wasmosy case, Oviedo remained a political refugee in Brazil until 2004, when he returned and was convicted.

The Supreme Court later exonerated Oviedo after military officers denied there had been a coup attempt, freeing him to run for president in 2008. He came in third, splitting the vote that gave former Catholic bishop Fernando Lugo the presidency and ended 60 years of one-party rule by the Colorado Party.

Oviedo was born in the small village of Juan de Mena, outside Asuncion, on Sept. 23, 1943. He graduated from the nation's military academy, and married Raquel Marin, an Argentine, with whom he had three children. Oviedo also had other children with two Paraguayan women before his marriage.

Oviedo's family now maintains his legacy in his UNACE party: His children Fabiola and Ariel are deputies in Congress, and his nephew, Lino Cesar Oviedo Sanchez, is a senator.

Oviedo "had an enormous capacity for work. At 5 a.m. he was in his office, receiving people until midnight, usually the poor, who came to him from their villages seeking help," former Sen. Enrique Gonzalez, a longtime political ally, told The Associated Press.

"He was an individual with great charisma, he spoke (Paraguay's indigenous language of) Guaran? perfectly, and he wove jokes in Guarani into his speeches. He had the spiritual strength to put up with being persecuted. His military preparation enabled him to put up with extreme situations. In December, he even managed to come out of a Brazilian hospital with two stents after a heart operation with more enthusiasm than ever."

U.S. Undersecretary of State Peter Romero had declared after the Wasmosy affair that Oviedo "lacks democratic credentials."

Gonzalez took issue with that label, noting that Oviedo also "founded a political party and participated in elections, building it into Paraguay's third-largest political force."

(AP) ? The fate of his gun proposals on Capitol Hill uncertain, President Barack Obama is seeking to rally support from the public and law enforcement community for his calls to ban assault weapons and install universal background checks for gun buyers.

Obama will pitch his proposals to stem gun violence Monday in Minnesota, a Democratic-leaning state where officials have been studying ways to reduce gun-related attacks and accidents for several years. His visit to the Minneapolis Police Department's Special Operations Center will mark the first time Obama has campaigned on his controversial proposals outside of Washington.

Ahead of the trip, the White House released a photo of the president skeet shooting at Camp David, the presidential retreat. Obama cited skeet shooting when asked in a recent interview whether he had ever shot a gun.

The president unveiled his sweeping package of proposals for curbing gun violence last month in response to the horrific mass shooting at a Newtown, Conn., elementary school. He vowed to use the full weight of his office to fight for the proposals, many of which face tough opposition from congressional lawmakers and the powerful National Rifle Association.

The reinstatement of the assault weapons ban, which expired in 2004, is expected to be the steepest climb for Obama. Universal background checks for gun purchasers may have an easier time passing Congress, though the NRA also opposes that measure.

Senate Judiciary Chairman Patrick Leahy, D-Vt., has said he hopes his panel can write gun legislation this month, though it's unclear what it will contain.

The White House picked Minneapolis as the backdrop for Obama's remarks in part because of recent steps the city has taken to tackle gun violence, including a push for stricter background checks.

After a spike in violent crimes, the city launched a program in 2008 aimed at providing more resources for at-risk youth and helping rehabilitate young people who have already perpetrated crimes. In January, Minneapolis also hosted a regional summit on gun violence for elected officials from around the Midwest.

Minneapolis Police Chief Janee Harteau and Hennepin County Sheriff Richard Stanek are also among the officials Obama has consulted as he pursues his anti-gun violence measures.

Stanek has also been leading a group of Minnesota sheriffs pushing for stronger background checks for people trying to buy guns.

Obama is expected to make more trips around the country to build support for his anti-gun violence measures. The outside group Organizing For Action, an offshoot of Obama's presidential campaign, is also promoting the proposals.

White House officials say quick action on the president's gun measures gives them the best prospects for passing legislation in Congress. They fear that as time passes lawmakers will have less incentive to back the measures as the shock of the Newtown massacre fades.

In addition to the gun control measures, Obama's anti-violence proposals also included increasing mental health resources, boosting funding for school security, and lifting restrictions that prevent the government from studying the causes of gun violence.

Mai doesn?t hit the right chords

What Alia Bhatt attempted at 19, Asha Bhosle does at 79 in ?Mai?. But that?s where the similarities between the AB debutantes ends. Mahesh Kodiyal?s film feels like old wine in an older bottle and even though Bhosle?s performance isn?t godawful, this film hardly ever hits the right chords.

Madhu (Padmini Kolhapure) and her husband Subhash Joshi?s (Ram Kapoor) family life is disrupted when her brother Munna decides to move to the United States for three years. Refusing Munna?s suggestion that they send their mother (Asha Bhosle) to an old age home, Madhu decides to bring her to her house instead. With no support from her husband and daughter about the new arrangement, Madhu?s woes increase when she finds out that her Mai?s forgetfulness is due to Alzheimer?s disease. Things get from bad to worse when Mai forgets how to wear her sari or when she sits down for a meal right after a heavy dinner. While Subhash, a journalist receives recognition for his work, Madhu refuses a promotion to take care of her mother.

Loosely inspired by ?Bhagban?, ?Mai? starts off as a story on the done-to-death topic of how children should (but don?t) take care of their parents. A teenage granddaughter who doesn?t like making adjustments, (there is even a song with lyrics like ?learn to adjust?) predictably turns over a new leaf just like in Amitabh Bachchan and Hema Malini-starrer.

To be fair, Asha Bhosle does a decent job ? far better than other singers who?ve recently turned actors but it?s hardly a role worth boasting about (here?s looking at you Bappida). Although some of her scenes as an Alzheimer?s patient are touching, the badly written dialogues and shoddy set ups spoils the mood. Padmini Kolhapure and Ram Kapoor also hold up the fort in the acting department, but the stale concept drowns out their best efforts.

Listen to an old Asha Bhosle melody instead of watching ?Mai? and keep your love for this legend intact.

WASHINGTON (AP) ? Goodbye candy bars and sugary cookies. Hello baked chips and diet sodas.

The government for the first time is proposing broad new standards to make sure all foods sold in schools are more healthful, a change that would ban the sale of almost all candy, high-calorie sports drinks and greasy foods on campus.

Under new rules the Department of Agriculture proposed Friday, school vending machines would start selling water, lower-calorie sports drinks, diet sodas and baked chips instead. Lunchrooms that now sell fatty "a la carte" items like mozzarella sticks and nachos would have to switch to healthier pizzas, low-fat hamburgers, fruit cups and yogurt.

The rules, required under a child nutrition law passed by Congress in 2010, are part of the government's effort to combat childhood obesity. While many schools already have made improvements in their lunch menus and vending machine choices, others still are selling high-fat, high-calorie foods.

Under the proposal, the Agriculture Department would set fat, calorie, sugar and sodium limits on almost all foods sold in schools. Current standards already regulate the nutritional content of school breakfasts and lunches that are subsidized by the federal government, but most lunch rooms also have "a la carte" lines that sell other foods. And food sold through vending machines and in other ways outside the lunchroom has not been federally regulated.

"Parents and teachers work hard to instill healthy eating habits in our kids, and these efforts should be supported when kids walk through the schoolhouse door," said Agriculture Secretary Tom Vilsack.

Most snacks sold in school would have to have less than 200 calories. Elementary and middle schools could sell only water, low-fat milk or 100 percent fruit or vegetable juice. High schools could sell some sports drinks, diet sodas and iced teas, but the calories would be limited. Drinks would be limited to 12-ounce portions in middle schools, and 8-ounce portions in elementary schools.

The standards will cover vending machines, the "a la carte" lunch lines, snack bars and any other foods regularly sold around school. They would not apply to in-school fundraisers or bake sales, though states have the power to regulate them. The new guidelines also would not apply to after-school concessions at school games or theater events, goodies brought from home for classroom celebrations, or anything students bring for their own personal consumption.

The new rules are the latest in a long list of changes designed to make foods served in schools more healthful and accessible. Nutritional guidelines for the subsidized lunches were revised last year and put in place last fall. The 2010 child nutrition law also provided more money for schools to serve free and reduced-cost lunches and required more meals to be served to hungry kids.

Iowa Sen. Tom Harkin, a Democrat, has been working for two decades to take junk foods out of schools. He calls the availability of unhealthful foods around campus a "loophole" that undermines the taxpayer money that helps pay for the healthier subsidized lunches.

"USDA's proposed nutrition standards are a critical step in closing that loophole and in ensuring that our schools are places that nurture not just the minds of American children but their bodies as well," Harkin said.

Last year's rules faced criticism from some conservatives, including some Republicans in Congress, who said the government shouldn't be telling kids what to eat. Mindful of that backlash, the Agriculture Department exempted in-school fundraisers from federal regulation and proposed different options for some parts of the rule, including the calorie limits for drinks in high schools, which would be limited to either 60 calories or 75 calories in a 12-ounce portion.

The department also has shown a willingness to work with schools to resolve complaints that some new requirements are hard to meet. Last year, for example, the government relaxed some limits on meats and grains in subsidized lunches after school nutritionists said they weren't working.

Schools, the food industry, interest groups and other critics or supporters of the new proposal will have 60 days to comment and suggest changes. A final rule could be in place as soon as the 2014 school year.

Margo Wootan, a nutrition lobbyist for the Center for Science in the Public Interest, says surveys done by her organization show that most parents want changes in the lunchroom.

"Parents aren't going to have to worry that kids are using their lunch money to buy candy bars and a Gatorade instead of a healthy school lunch," she said.

The food industry has been onboard with many of the changes, and several companies worked with Congress on the child nutrition law two years ago. Major beverage companies have already agreed to take the most caloric sodas out of schools. But those same companies, including Coca-Cola and PepsiCo, also sell many of the non-soda options, like sports drinks, and have lobbied to keep them in vending machines.

A spokeswoman for the American Beverage Association, which represents the soda companies, says they already have greatly reduced the number of calories kids are consuming at school by pulling out the high-calorie sodas.
